Grillo was born in New York City, New York. He is probably best known for his role as Nick Savrinn in Season One of Prison Break. He also had roles in other television shows and movies, including the short-lived television series Blind Justice and Battery Park. He is also known for his very popular stint as Hart Jessup on the daytime drama Guiding Light from May 1996 through March 1999.

Recently, Frank Grillo played "Mr. Pig" in the Spike TV Original Drama Series, "The Kill Point" which aired during the summer of 2007. He also played the role of Jimmy in the CSI New York episode "The Things About Heroes (CSI: NY episode)", which aired on the 28th November 2007. His full frontal nude black and white photo shoot remains popular.

He married his first wife Kathy in 1991, but the couple divorced in 1998. They had a son, Remy (born in January 1997).

Grillo is married to fellow actress Wendy Moniz, also of Portuguese descent. The couple has three boys Liam (born August 2004) and twins Rio Joseph Grillo and Dylan Jade Grillo (born January 25, 2008).
